Lipopolysaccharide may change how ligament stem cells develop and affect immune response through ERK1/2 signaling

Abstract

Lipopolysaccharide (LPS) treatment altered the differentiation capacity of human periodontal ligament stem cells (PDLSCs).

  • LPS did not affect the immunophenotype, proliferation, viability, or cell cycle of PDLSCs.
  • LPS inhibited osteogenesis by decreasing mRNA expression of Runx2, ALP, and Ocn while promoting chondrogenesis and adipogenesis through increased Sox9 and PPARγ mRNA expression.
  • LPS enhanced the contractility of PDLSCs and increased the expression of proteins and genes related to myofibroblast characteristics, including TGF-β, fibronectin, α-SMA, and NG2.
  • LPS elevated the levels of anti-inflammatory COX-2 and pro-inflammatory IL-6 in PDLSCs.
  • LPS-treated PDLSCs reduced the transendothelial migration of peripheral blood mononuclear cells (MNCs) and altered CD29 expression in MNCs.
