Lipid Nanoparticle Delivery of CF10 Boosts Effectiveness Against Colorectal Cancer Cells While Keeping Dual Targeting of Tumor Suppressor and Topoisomerase 1

Abstract

Lipid nanoparticles (LNPs) formed with the nanoscale DNA-based polymer CF10 display a size range of 80-200 nm and a zeta-potential of -15 mV.

  • LNPs showed stability in PBS over a 4-week period.
  • In vitro studies indicated that LNP formulation increased uptake of CF10 specifically in cancer cells, while nonmalignant cells did not show increased uptake.
  • CF10:LNPs primarily colocalized with lysosomes after 48 hours, following initial colocalization with endosomes.
  • Increased cytotoxicity to cancer cells was observed with CF10:LNPs compared to free CF10, correlating with higher cell uptake.
  • Efficient release of active nucleotides and potent inhibition of thymidylate synthase were demonstrated with CF10:LNPs, maintaining dual targeting of thymidylate synthase and DNA topoisomerase 1.
