A long-term cell model for studying aging-related changes in immune cells

Updated

Abstract

Essence

Long-term culture pushed human macrophages toward senescence-associated proinflammatory states with reduced cytokine flexibility.

Evidence

This in vitro study cultured human monocyte-derived macrophages for 7, 14, or 21 days and measured senescence markers, flow-cytometry phenotypes, and LPS-stimulated cytokine responses.

Caveat

Because this is a cell-culture model, its implications for immune aging and inflammation in living humans remain preclinical.

What this is

  • This research investigates macrophage senescence, a process linked to immune dysfunction in aging.
  • A long-term culture model was developed to study the phenotypic and functional changes in human macrophages over time.
  • Monocyte-derived macrophages were cultured for 7, 14, and 21 days, revealing progressive senescence markers and altered immune responses.

Essence

  • Extended culture of human macrophages induces a senescent phenotype characterized by altered marker expression and cytokine profiles. This model aids in understanding immune aging and potential therapeutic strategies.

Key takeaways

  • Macrophages cultured for 21 days exhibited significant upregulation of senescence markers, including CDKN2A (p16INK4a) and β-galactosidase. This indicates a clear progression toward a senescent state.
  • Cytokine secretion profiles changed dramatically; day 7 macrophages produced a diverse array of cytokines, while day 21 macrophages showed reduced secretion of Th1-type cytokines, reflecting a dysregulated immune response.
  • High-dimensional flow cytometry revealed five distinct macrophage subpopulations, with a shift from anti-inflammatory to proinflammatory phenotypes as cultures aged, indicating loss of plasticity and functional reprogramming.

Caveats

  • The model's physiological relevance may be limited by the absence of environmental cues and donor variability, which could affect the generalizability of findings.
  • Future studies should assess phagocytic capacity and reactive oxygen species production to provide a more comprehensive understanding of macrophage functionality in senescence.

Definitions

  • senescence-associated secretory phenotype (SASP): A phenotype characterized by the secretion of proinflammatory cytokines and other factors by senescent cells, contributing to chronic inflammation.
