Long-term immunity and protection against SARS-CoV-2 XBB.1.5 after matching or mixed mRNA and MVA vaccines

Updated

Abstract

Binding IgG antibodies against the XBB.1.5 spike protein remained high throughout the 9-month period in all vaccinated groups.

  • Mice received homologous and heterologous vaccination regimens using either an mRNA vaccine or an MVA-based candidate.
  • Neutralizing antibody levels peaked shortly after vaccination and gradually declined over time, converging across different regimens.
  • SARS-CoV-2-specific CD8+ T-cell responses were strongest in mRNA-containing groups at day 10 and remained detectable at 9 months.
  • All two-dose regimens significantly lowered viral RNA levels and infectious viral titers after challenge with SARS-CoV-2 XBB.1.5.
  • Vaccinated mice showed reduced expression of genes related to inflammation and cellular stress in lung tissue compared to controls.
  • Distinct transcriptional profiles were identified in lungs of mice, reflecting variations in immune responses among vaccination regimens.
