Loop shape in mRNA vaccine tail improves protein production and mRNA stability

Abstract

Essence

Adding a loop structure to an mRNA poly(A) tail improved expression and stability but did not clearly strengthen immune responses.

Evidence

In vitro and mouse mRNA platform experiments measured bioluminescence, in vivo hEPO expression, and HPV or influenza antigen immune responses across different poly(A) tail structures.

Caveat

The work is preclinical, and the higher expression did not produce significant differences in T cell immunity or antibody titers for the tested antigens.
