Low-dose bone growth protein and stem cell delivery on coral scaffold for healing large bone defects in sheep

Abstract

Only 50% of the bone defects healed in sheep using tissue-engineered constructs with mesenchymal stem cells and low-dose bone morphogenetic protein-2.

  • Bone union with nearly complete scaffold resorption was observed in 3 out of 6 animals with MSC-BMP constructs.
  • 2 out of 7 animals showed healing with MSC-only constructs, and 1 out of 5 healed with BMP-only constructs.
  • The amount of newly formed bone did not significantly differ among the BMP, MSC, and MSC-BMP groups.
  • Increased scaffold resorption rate was noted with BMP-TECs as a potential side effect.
  • Further refinement of this tissue-engineering strategy is necessary before clinical application.
