Lung surfactants in fat-based nanoparticles for delivering mRNA to the lungs

Abstract

Essence

Adding Poractant alfa lung surfactant may improve lipid nanoparticle assembly and pulmonary mRNA delivery.

Evidence

This formulation and preclinical experiment tested surfactant-enhanced mRNA lipid nanoparticles in vitro and after intranasal administration in mice, reporting higher particle concentration, increased transfection and protein expression, cytocompatibility, and stronger lung mCherry expression than conventional LNPs.

Caveat

The evidence is limited to cell and mouse experiments with reporter expression, not therapeutic efficacy or human pulmonary delivery.
