Lung surfactants in lipid nanoparticles for delivering mRNA to the lungs

Updated

Abstract

Lung surfactant-enhanced lipid nanoparticles (Surf-LNPs) showed a 10-fold increase in particle concentration compared to conventional formulations.

  • Surf-LNPs demonstrated several-fold increases in mRNA transfection efficiency and protein expression in vitro.
  • The incorporation of surfactant protein B (SP-B) is associated with more rapid and efficient endosomal escape compared to conventional LNPs.
  • In vivo studies indicated enhanced mCherry expression in the lungs of mice treated with Surf-LNPs following intranasal administration.
  • These findings support the potential of lung surfactant incorporation to improve the effectiveness of pulmonary gene delivery systems.
