Abstract

Lung-targeted lipid nanoparticles significantly prolonged survival in orthotopic models of Non-Small-Cell Lung Cancer (NSCLC).

  • Lung-enriched gamma delta T cells are identified as a critical component in NSCLC.
  • Analysis of patient data indicates that gamma delta T cell and CD1d signatures correlate with improved survival in NSCLC patients.
  • The engineered lipid nanoparticles preferentially accumulate in the lung after intravenous administration.
  • Activation of gamma delta T cells and enhanced CD8+ T cell infiltration were observed in NSCLC models treated with the nanovaccine.
  • Depletion of gamma delta T cells eliminated the therapeutic benefits, highlighting their role as essential effector cells.
  • Splenectomy reduced the effectiveness of the vaccine, indicating the importance of systemic immune interactions.
