Lysosome Problems Linked to Spinal Disc Wear: Molecular Types and Key Genes Found Using Advanced Analysis

Two lysosome-related molecular subtypes were identified in intervertebral disc degeneration (IVDD).

  • A senescence/inflammation-enriched subtype and a metabolism-enriched subtype were found.
  • The lysosomal gene-signature score is positively associated with the senescence score, indicating coordinated transcriptional activation.
  • PLD3 was identified as a candidate hub gene, with its knockdown worsening lysosomal impairment and senescence in treated cells.
  • Lithocholic acid partially improved lysosomal function and reduced degeneration-associated changes in vitro and in a rat model.
