Fixing lysosome problems reduces cell aging in Hutchinson-Gilford progeria syndrome

Updated

Abstract

Primary cells from HGPS patients exhibited lysosome defects that hindered progerin clearance.

  • Approximately 90% of HGPS cases are linked to mutations in the LMNA gene, producing the toxic protein progerin.
  • Lysosome defects in HGPS cells impair the clearance of progerin, contributing to cellular senescence.
  • Activating lysosome biogenesis through specific compounds enhanced progerin clearance and reduced signs of cellular aging.
  • Progerin was expelled from the nucleus into the cytoplasm and degraded via a process involving autophagy.
  • The enhancement of lysosome function was associated with decreased DNA damage, improved cell cycle progression, and reduced senescence markers.
