Macrophage Fat Balance May Contribute to Spinal Disc Degeneration by Reducing Clearance of Aging Cells

Abstract

Advanced intervertebral disc degeneration (IVDD) is associated with increased disc fat fraction and systemic lipid abnormalities.

  • Lipid metabolic disturbances may contribute to immune remodelling in degenerative discs.
  • Four hub genes linked to lipid metabolism were identified: TIAM2, SLC44A4, PPT1, and PTGDS.
  • Disc macrophages with low glycerophospholipid metabolism scores may exhibit higher senescence activity and inflammatory polarization.
  • Lipid overload in macrophages could lead to lipid droplet accumulation and activation of the p53/p21 senescence pathway.
  • Restoring lipid homeostasis with rosuvastatin may improve macrophage function and reduce inflammatory markers in the disc.
