Single-cell RNA sequencing revealed a reduced proportion of peritoneal mesothelial cells and an increased number of fibroblasts in long-term peritoneal dialysis patients.

  • Long-term peritoneal dialysis is associated with activated senescence pathways in peritoneal mesothelial cells.
  • High glucose environments trigger senescence in peritoneal mesothelial cells, which may contribute to peritoneal fibrosis.
  • The transcription factor KLF6 enhances RRAS expression, which is involved in the senescence process of mesothelial cells.
  • Senescent mesothelial cells activate fibroblasts through the secretion of TGF-β, leading to increased fibrosis markers in fibroblasts.
  • KLF6 knockdown and TGF-β receptor inhibition can block the activation of fibroblasts induced by senescent mesothelial cells.
