How gut bacteria break down bile acids through a metabolic process

Abstract

A set of six enzymes is necessary and sufficient for the eight-step conversion of cholic acid to deoxycholic acid (DCA) and lithocholic acid (LCA).

  • The gut microbiota produce secondary bile acids like DCA and LCA, which can affect host physiology.
  • DCA and LCA are associated with blocking the growth of Clostridium difficile and promoting hepatocellular carcinoma.
  • The pathway to synthesize DCA and LCA has been completed by characterizing the enzymes involved.
  • Engineering the pathway into Clostridium sporogenes allows for the production of DCA and LCA in a nonproducing bacterium.
  • This work demonstrates the potential to express and control microbiome-derived pathways heterologously.
