Metformin partly reduces nerve cell aging caused by cisplatin but does not stop increased sensitivity in rat peripheral nerves

Updated

Abstract

Metformin showed partial senomorphic activity in counteracting cisplatin-induced senescence, but did not significantly alleviate chemotherapy-induced peripheral neuropathy.

  • Cisplatin exposure in motor neurons induced features of senescence, such as increased senescence-associated β-galactosidase activity and upregulation of senescence markers.
  • Metformin treatment attenuated the expression of key senescence-associated transcripts, including CDKN2A and components of the senescence-associated secretory phenotype.
  • In a rat model, cisplatin administration caused mechanical allodynia and thermal hyperalgesia, alongside strong senescence markers in dorsal root ganglia.
  • Metformin pre-treatment reversed the upregulation of senescence markers in the rats but did not improve sensory deficits caused by cisplatin.
