Astrocyte problems linked to MFN2 may lead to overactive nerve cells and depression-like behaviors

Updated

Abstract

Astrocytic mitofusin-2 (MFN2) expression is elevated in patients with major depressive disorder (MDD) and is associated with astrocyte dysfunction.

  • Increased expression of Mfn2 was observed in plasma, postmortem brain tissue, and patient-derived astrocytes linked to MDD.
  • Chronic social defeat stress (CSDS) in mice resulted in up-regulation of Mfn2, excessive mitochondrial fusion, impaired mitophagy, and loss of astrocytes.
  • Knockdown of Mfn2 in astrocytes improved mitochondrial structure and function, preserving astrocyte health and reducing depressive-like symptoms in mice.
  • Reduced stability of the transcription factor STAT1, due to β-arrestin2 deficiency, was linked to the increased expression of Mfn2.
  • Decreasing MFN2 levels or enhancing STAT1 expression led to lower extracellular glutamate levels and mitigated neuronal hyperexcitability in stressed mice.
