Mfn2-linked mitochondrial problems may cause kidney scarring by triggering cell death in chronic kidney disease

Abstract

Expression of glutathione peroxidase 4 (GPX4) and Parkin was decreased in the renal tissues of chronic kidney disease patients.

  • Reduced levels of GPX4 and Parkin were negatively correlated with serum creatinine and C-reactive protein.
  • These protein levels were positively correlated with estimated glomerular filtration rate.
  • In mouse models and human kidney cells, mitophagy-related proteins were found to be decreased in renal fibrosis.
  • The mitochondrial uncoupler CCCP reduced renal fibrosis by inhibiting ferroptosis, while mitochondrial division inhibitor 1 (Mdivi-1) had the opposite effect.
  • Mitofusin2 (Mfn2) was identified as a key player in both mitophagy and ferroptosis.
  • Knockdown of Mfn2 inhibited mitophagy and worsened ferroptosis and renal fibrosis, while its overexpression improved these conditions.
