Mitochondrial connexin 43 helps glioma cells adjust to metabolic stress

Abstract

Basal CX43 expression and extracellular stress factors significantly modulated the mitochondrial localisation of CX43.

  • Mitochondrial CX43 () expression and localization were influenced by cell density and extracellular pH fluctuations.
  • Inhibition of the heat shock protein 90 (HSP90) chaperone system led to a marked reduction in mt-CX43 levels.
  • Geldanamycin treatment increased purine biosynthesis in T98-G cells, associated with lower basal CX43 and mt-CX43 under stress.
  • U-87 MG cells showed a stable NAD/NADH ratio and increased NADH levels, suggesting a metabolic shift towards resilience.
  • mt-CX43 may play a role in regulating metabolic adaptation and stress responses in glioma cells.
