Mitochondrial DNA mutations linked to weaker energy production and better imatinib response in chronic myeloid leukemia

Abstract

A total of 241 somatic mitochondrial DNA mutations were identified in 92 patients with chronic myeloid leukemia.

  • Distinct mitochondrial DNA mutational patterns are associated with different metabolic programs in leukemic stem cells.
  • Patients with chronic myeloid leukemia who had three or more mitochondrial DNA mutations showed better responses to imatinib treatment.
  • Higher mitochondrial DNA mutation frequency correlated with reduced respiratory reserve and increased mitophagy in favorable responders.
  • Suboptimal responders demonstrated enhanced mitochondrial respiration and lipid metabolic activity.
  • Pharmacologic inhibition of Complex I decreased clonogenic potential and increased sensitivity to imatinib.
