Mitochondrial Problems Linked to α-Synuclein Toxicity in Lab Models of Protein Clumps

Updated

Abstract

33 peer-reviewed studies were included in a systematic review examining the effects of misfolded α-synuclein on mitochondrial function.

  • Oligomeric and fibrillar α-synuclein consistently induced mitochondrial depolarisation and impaired oxidative phosphorylation.
  • These forms of α-synuclein reduced ATP production and caused dysfunction in complex I of the mitochondrial respiratory chain.
  • Increased generation of reactive oxygen species and mitochondrial fragmentation were observed alongside disrupted cristae and impaired protein import.
  • Calcium dysregulation, defective mitophagy, and accumulation of damaged mitochondria were also noted as mitochondrial pathological changes.
  • Mitochondrial dysfunction often preceded cell death and was more pronounced in dopaminergic and genetically susceptible models.
