Mitochondrial dysfunction is increasingly recognized as a pivotal mechanism in diabetic retinopathy (DR).

  • Long-standing hyperglycemia and metabolic dysregulation are associated with retinal neurovascular unit injury.
  • Excessive glucose flux under hyperglycemic conditions leads to mitochondrial reactive oxygen species (ROS) overproduction and mitochondrial DNA (mtDNA) damage.
  • Impaired oxidative phosphorylation and mitochondrial dynamics contribute to ATP depletion and inflammation.
  • Mitochondrial injury affects various retinal cell types, which may disrupt the blood-retinal barrier and contribute to neurodegeneration.
  • Emerging therapeutic strategies targeting mitochondrial dysfunction could provide new avenues for DR prevention and treatment.
