How the human mitochondrial processing enzyme identifies and processes PINK1 protein

Abstract

The mitochondrial processing peptidase (MPP) cleavage site on PINK1 is defined between Ala28 and Tyr29.

  • PINK1 is inefficiently processed by MPP compared to typical mitochondrial targeting sequences.
  • MPP cleavage is not necessary for PARL processing or PINK1 function in cellular contexts.
  • In vitro, the PINK1 N-terminal targeting sequence binds strongly to MPP, inhibiting the cleavage of other substrates.
  • This binding traps MPP in a slowly processing complex, affecting its overall activity.
  • A two-step mechanism for MPP processing involving rearrangement of the MPPα lid and active site engagement was identified.
