Brain cell damage in rats from 2,5-hexanedione linked to a process where damaged mitochondria trigger iron-related cell death

Abstract

Chronic exposure to n-hexane's metabolite, 2,5-hexanedione, leads to significant neuronal damage in rats.

  • HD exposure results in iron accumulation, lipid peroxidation, and mitochondrial abnormalities in spinal neurons.
  • Treatment with the iron chelator deferoxamine or the ferroptosis inhibitor Ferrostatin-1 helps preserve mitochondrial integrity.
  • Activation of mitophagy is indicated by increased levels of Beclin-1, LC3II, Drp-1, and PINK-1 in spinal mitochondria following HD exposure.
  • Inhibition of mitophagy with chloroquine reduces iron deposition and lipid peroxidation, while improving motor function.
  • Knockdown of PINK-1 in human neuroblastoma cells reduces mitophagy and ferroptosis induced by HD.
