Low levels of cell cleanup markers link to harmful oxygen buildup in cows with fatty liver and cause oxygen buildup and fat buildup in calf liver cells

Abstract

Serum concentrations of β-hydroxybutyrate were higher in dairy cows with fatty liver.

  • Hepatic levels of malondialdehyde (MDA) and hydrogen peroxide (HO) were elevated in cows with fatty liver.
  • Reduced protein levels of PINK1, Parkin, p62, and LC3-II in the liver indicate impaired mitophagy in dairy cows with fatty liver.
  • Knockdown of the PRKN gene in hepatocytes led to decreased p62 and LC3-II levels, while increasing triacylglycerol (TG), MDA, and HO levels.
  • Higher concentrations of free fatty acids (FFA) resulted in lower protein levels of mitophagy markers and increased lipid and oxidative stress markers in hepatocytes.
  • Overexpression of PRKN in FFA-treated hepatocytes enhanced mitophagy marker levels and reduced lipid and oxidative stress markers.
