Importance of mitochondrial cleanup in nerve cell death caused by reactive oxygen linked to pyrrolidinophenones

Updated

Abstract

Treatment with α-pyrrolidinooctanophenone (POP) resulted in significant neuronal cell toxicity.

  • Neuronal toxicity from pyrrolidinophenones (PPs) varies with the length of the alkyl chain.
  • PPs exposure led to increased production of reactive oxygen species (ROS) in human neuronal cells.
  • Antioxidant treatment with N-acetyl-L-cysteine reduced the cytotoxic effects of PPs, indicating ROS's role in toxicity.
  • PPs accumulated in mitochondria and activated pro-apoptotic signals, evidenced by changes in the Bax/Bcl2 ratio and caspase-9 activation.
  • POP induced mitophagy through the PINK1/Parkin pathway, which appears to protect against ROS-induced damage.
  • Inhibiting autophagy or mitophagy increased ROS production and cell death, suggesting mitophagy's role in mitochondrial quality control.
