MMP14 and FOS as key controllers of blood vessel cell aging in diabetic kidney disease

Abstract

43 candidate genes were identified as potentially regulating vascular smooth muscle cell senescence in diabetic kidney disease.

  • Key genes showed enrichment in biological aging, cellular senescence, oxidative damage, insulin signaling, and MAPK pathways.
  • MMP14 and FOS were identified as pivotal genes with potential diagnostic capabilities.
  • Immune cell composition analysis indicated significant changes in immune populations correlated with MMP14 and FOS expression.
  • Computational predictions highlighted MMP14, FOS, NEAT1, and hsa-miR-181a-5p as critical regulatory nodes in the ceRNA network.
  • Five therapeutic candidates were identified, with linsitinib showing the most favorable binding affinity in simulations.
  • Under hyperglycemic conditions, HASMCs displayed reduced proliferation and increased apoptosis and senescence, which were influenced by MMP14 and FOS expression.
