Molecular details of a clumping-prone alpha-synuclein variant used to model brain diseases with protein buildup

Updated

Abstract

The artificial variant SynT shows increased aggregation propensity compared to normal alpha-synuclein.

  • SynT was structurally characterized using Nuclear Magnetic Resonance (NMR) spectroscopy.
  • Conformations of SynT resemble those of unmodified alpha-synuclein, supporting its use as an aggregation model.
  • Differences in the N-terminal region of SynT involve interactions that may influence alpha-synuclein aggregation.
  • Decreased membrane binding of SynT is associated with its aggregation behavior.
