Monomethyl fumarate (MMF) increased MTT reduction in a dose-dependent manner.

  • Dimethyl fumarate (DMF) showed a biphasic response, enhancing MTT reduction at low concentrations but inducing toxicity at higher levels.
  • Increased MTT reduction with MMF reflects enhanced cellular metabolic activity, not increased cell viability.
  • MMF treatment led to higher oxygen consumption and ATP production, indicating improved mitochondrial function.
  • Proteomic analysis suggested a qualitative shift in mitochondrial homeostasis rather than an upregulation of respiratory chain proteins.
  • MMF influenced microglial iron metabolism, reducing the uptake of toxic non-transferrin-bound iron.
  • A protective response to iron overload was indicated by the upregulation of the ferroptosis suppressor protein.
