Delivering mRNA to joint cells using polymer nanoparticles in lab and living systems

Updated

Abstract

Poly(amidoamine)-based nanoparticles (PAA-based NPs) showed higher luciferase expression in osteoarthritis (OA) rat knee joints compared to healthy joints.

  • Cationic uncoated NPs demonstrated prolonged retention in OA rat knee joints.
  • PEG-coated NPs exhibited higher cell uptake and equivalent gene expression in 2D chondrocyte cultures.
  • Both NP types penetrated cartilage and were taken up by chondrocytes in ex vivo joint cultures.
  • Intra-articular administration did not affect cartilage integrity in healthy or OA rat joints, but increased synovial inflammation was observed.
  • The effectiveness of mRNA delivery may depend on the properties of the nanoparticles and the disease status of the joint.
