mRNA Nanoparticles Producing a Key Bacterial Protein Trigger Wide-Ranging Systemic and Mucosal Antibodies Against Drug-Resistant Gonorrhea

Abstract

BamA mRNA-lipid nanoparticles elicited robust antibodies against Neisseria gonorrhoeae in a female mouse model.

  • Both intramuscular and intranasal delivery routes produced strong BamA-specific antibody responses.
  • Antibodies recognized a variety of Neisseria gonorrhoeae isolates, including those resistant to ceftriaxone.
  • Neither vaccination route led to quicker bacterial clearance or reduced bacterial load.
  • The formulations did not generate serum bactericidal activity.
  • While immunogenic, the current BamA mRNA-LNP vaccine does not provide protection, suggesting the need for further modifications.
