How mRNA-Lipid Nanoparticles Help Gene Editing and Affect Immune Responses

Updated

Abstract

Up to 80% gene editing efficiency has been achieved in vivo using a nonviral lipid nanoparticle approach for CRISPR-Cas9.

  • mRNA plays a crucial role in protein synthesis, impacting gene editing and therapies.
  • MEVPCS-mRNA lipid nanoparticles elicit strong immune responses comparable to existing HIV vaccines.
  • CD8+ T cell responses were significantly triggered by lipid nanoparticles carrying mRNA.
  • In mice immunized with LNP-mRNA, notable Th1-type CD4+ T cell responses against Mycobacterium tuberculosis were observed.
  • Variations in pH, ionic strength, and freezing rate are important for improving the preservation of mRNA lipid nanoparticles.
  • Structural degradation of lipid nanoparticles was indicated by an increase in particle size from 85 nm to 300 nm.
