mRNA nanoparticle delivery of linked nanobodies protects young geese from new goose virus infection

Updated

Abstract

The 3×Nb60 mRNA-LNP treatment resulted in 100% survival of infected goslings at a dosage of 2 mg/kg.

  • A neutralizing nanobody (Nb60) targeting the spike protein of novel goose astrovirus (nGAstV) effectively inhibits viral replication.
  • The mRNA-encoded homo-tandem nanobody (3×Nb60) demonstrated approximately twofold improvement in neutralization potency compared to the Nb60 monomer.
  • Lipid nanoparticle encapsulation of the mRNA enhances its serum half-life by approximately 48 hours.
  • Treatment with 3×Nb60 significantly reduced viral shedding and loads in tissues while restoring serum levels of uric acid and liver enzymes.
  • Epitope mapping identified critical binding residues on the P2 protein, suggesting a specific target for the nanobody.
