mRNA Vaccines with Optimized Dengue Proteins Provide Protection Without Causing Harmful Immune Response

Updated

Abstract

Essence

Dengue mRNA-LNP vaccines with fusion-loop changes protected mice while lowering antibody-dependent enhancement signals.

Evidence

This mouse vaccine study tested DENV 2, 3, and 4 mRNA-LNP prM-E vaccines and a tetravalent formulation, measuring serotype-specific neutralizing antibodies, lethal homotypic challenge protection, and ADE in vaccinated mouse sera.

Caveat

The protection and reduced ADE evidence is preclinical mouse data, so human efficacy, durability, and safety against all dengue serotypes remain untested in the abstract.
