Measuring mRNA Tail Length and Composition in Real Time

Abstract

An online method was developed to determine mRNA poly(A) tail length and content using an immobilized ribonuclease cartridge coupled with hydrophilic interaction liquid chromatography.

  • The new approach improves throughput compared to traditional methods that require extensive sample processing.
  • Three hydrophilic interaction liquid chromatography columns were tested for their ability to separate poly(A) fragments ranging from 40 to 120 nucleotides.
  • Systematic studies showed that increasing the column temperature from 30 to 80 °C affected separation performance.
  • A bimodal poly(A) profile was observed for one catalog mRNA, indicating variability in poly(A) tail characteristics.
  • A proof-of-concept study demonstrated that the new method can effectively assess poly(A) tail content in mixtures of polyadenylated and nonpolyadenylated mRNAs.
