Optimized liposomes containing 4 mol% of a specific PEG-lipid enabled efficient in vivo expression of mRNA.

  • Varying PEG-lipid parameters, such as chain length and architecture, influences mRNA-liposome complex stability and delivery efficiency.
  • The optimal PEG-lipid was identified as linear PEG2000 conjugated to a C14-dialkylglycerol anchor via a carbamate linker.
  • This optimized formulation led to robust immune responses when delivering mRNA encoding specific viral proteins.
  • Enhanced mRNA delivery may improve the effectiveness of antiviral and antitumor vaccines.
