Abstract
Total nucleoside-5'-triphosphates (NTPs) input and magnesium concentration significantly influence the yield and purity of RNA produced by in vitro transcription.
- High RNA yield and integrity are associated with specific NTP input levels and magnesium concentrations.
- Low concentrations of double-stranded RNA (dsRNA) were achieved alongside high-quality RNA constructs.
- Different RNA sequences and lengths can be effectively produced using the proposed IVT design.
- The findings may help in creating a more cost-effective and efficient platform for RNA manufacturing.
