Constitutive mTORC1 activation in skeletal muscle does not impair systemic glucose homeostasis in mice, even under dietary stress.

  • Chronic mTORC1 activation increased signaling and altered autophagy markers in mouse skeletal muscle.
  • A western diet led to increased body and fat mass and impaired glucose tolerance, regardless of genotype.
  • Knockout of the Depdc5 gene had minimal effects on fasting glucose and insulin levels.
  • Female knockout mice exhibited a modest increase in weight gain and fasting glucose when on a western diet.
  • Mitochondrial respiration and content remained unchanged by either the knockout or the western diet.
  • Transcriptomic analysis showed upregulation of genes related to cell division and immune response due to the knockout.
