Phosphorylation of TFEB is dephosphorylated by muscarinic acetylcholine receptor M5 (M5R) while S6K1 and 4EBP1 are phosphorylated.

  • mTORC1 regulates diverse cellular processes by phosphorylating both canonical and non-canonical substrates.
  • Phosphorylation of non-canonical mTORC1 substrates can occur independently from canonical substrates.
  • GPCRs influence mTORC1 signaling differently for various substrates.
  • M5R promotes phosphorylation of S6K1 and 4EBP1 while dephosphorylating TFEB.
  • Activation of M5R leads to stimulation of protein synthesis without affecting lysosome biogenesis.
  • The findings suggest that GPCRs may selectively regulate anabolic and catabolic processes through mTORC1.
