Lipid nanoparticles that pass through lung mucus deliver two gene therapies together for idiopathic pulmonary fibrosis

Abstract

The DAS-lipid nanoparticle formulation improved lung accumulation of nucleic acid drugs by 3.5-fold compared to free drug delivery.

  • Pyroptosis mediated by gasdermin D is linked to disease advancement in idiopathic pulmonary fibrosis (IPF).
  • The serum amyloid P component is associated with anti-fibrotic properties.
  • DAS-LNP enabled enhanced mucus penetration and effective delivery of GSDMD-targeting siRNA and SAP-expressing plasmid DNA.
  • In a bleomycin-induced IPF mouse model, si/pD-DAS-LNP reduced pulmonary inflammation and pro-fibrotic factor levels.
  • The treatment improved lung function and survival rates in the mouse model without compromising safety.
