Multiple biological analyses show that TGF-β signals cause aging in gum stem cells

Abstract

Spatial transcriptomic profiling revealed enriched Tgfb1 expression in the periodontium.

  • TGF-β1 treatment in human periodontal ligament stem cells (PDLSCs) resulted in a senescent phenotype characterized by G2 phase cell cycle arrest and increased reactive oxygen species (ROS) accumulation.
  • TGF-β1 is associated with the silencing of the PRKAG2 gene through DNA methylation, leading to heightened ROS production and DNA damage.
  • ATM signaling activation was observed in PDLSCs following TGF-β1 treatment, linking oxidative stress to cellular senescence.
  • Mitigation of PDLSC senescence was achieved by inhibiting ROS with N-acetyl-l-cysteine or reversing PRKAG2 silencing with decitabine.
