Safety and Action of a Natural Injectable Treatment Targeting Skin Aging Pathways

Abstract

A total of 62 active compounds were identified from 11 materials in the injectable formulation Dong-An Pharmacopuncture (DAP) aimed at addressing skin aging.

  • Network pharmacology analysis revealed 225 compound-target interactions across 292 edges.
  • Protein-protein interaction analysis identified a network with 1,334 edges and 10 core targets, including TNF, IL6, and TP53.
  • Four major mechanistic axes were identified: regulation of inflammation and oxidative stress, hormonal skin homeostasis, tissue repair and ECM remodeling, and cellular senescence regulation.
  • A toxicity study in Sprague-Dawley rats showed no mortality or abnormal clinical signs after a single subcutaneous dose of DAP at 1.0 mL/head.
  • The findings support the need for further experimental validation of DAP's predicted mechanisms and safety.
