Peptide-lipid particles for targeted delivery of DNA and siRNA into breast cancer cells

Abstract

The peptide/lipid hybrid system achieved near complete knockdown of the Bcl-2 protein in breast cancer cells.

  • A multifunctional peptide-based delivery system was developed to enhance gene delivery specifically to breast cancer cells.
  • The combination of the peptide with a lipid formulation resulted in improved gene expression and silencing compared to either method used alone.
  • This system demonstrated more efficient gene knockdown compared to the leading commercial siRNA delivery agent Lipofectamine® RNAiMAX.
  • Delivery of BCL2 siRNA to MCF-7 cells led to significant inhibition of cell migration in a wound healing assay.
  • The findings suggest that this system may be an effective candidate for targeted gene therapy in breast cancer.
