Abstract

CRM1 selectively reduced cell viability in lung cancer cell lines by inducing distinct forms of cell death.

  • CRM1 induces paraptosis through downregulation of Alix and upregulation of ATF4 and CHOP.
  • This compound is associated with disruption of mitochondrial membrane potential and induction of endoplasmic reticulum stress.
  • Accumulation of reactive oxygen species (ROS) was observed alongside CRM1 treatment.
  • CRM1 also induces apoptosis, as shown by DNA fragmentation and increased levels of cleaved caspase-3 and Bax.
  • Autophagy was promoted by CRM1, indicated by increased expression of Atg7 and LC3-II and enhanced autophagosome formation.
  • Pharmacological studies suggest independent induction of apoptosis, paraptosis, and autophagy by CRM1.
