Drugs that affect NAD+ metabolism protect nerve fibers inside and outside the brain in a lab model of nerve injury

Updated

Abstract

Combined treatments preserved axon integrity for up to 7 days in culture.

  • Wallerian degeneration is linked to the depletion of the NAD+ enzyme NMNAT2 and subsequent activation of SARM1.
  • Inhibition of SARM1 and modulation of NAD+ metabolism may offer therapeutic strategies for protecting axons.
  • Targeted inhibition of DLK-JNK signaling alongside NAD+ metabolism modulation effectively reduced axonal degeneration.
  • The combination treatment suppressed SARM1 NADase and DLK-JNK activation, indicating a protective mechanism.
  • A therapeutic window of at least 24 hours was identified for the combination regimen, suggesting delayed treatment may still be effective.
