Abstract

Short-term exposure to nanosized diesel exhaust particles (nDEPs) induced cytotoxicity and DNA damage in umbilical cord-derived mesenchymal stem cells (UC-MSCs).

  • Exposure to nDEPs caused apoptosis, necrosis, and DNA adduct formation in UC-MSCs.
  • Short-term exposure led to DNA hypomethylation and changes in gene expression related to stress responses.
  • Prolonged exposure resulted in oxidative DNA damage-induced senescence and a senescence-associated secretory phenotype.
  • Both short- and long-term exposures altered the profiles of small extracellular vesicle (sEV) miRNAs, impacting genes linked to cell survival and differentiation.
  • Senescent sEVs from nDEPs-exposed UC-MSCs induced a senescent-like phenotype in recipient cells, reducing proliferation.
  • Knockdown of MYC and SIRT1, targets of altered sEV miRNAs, indicated potential genomic instability related to telomere shortening.
