How Removing Natural Killer Cells May Change Immune Responses to COVID mRNA Vaccines

Updated

Abstract

Primed NK cells increased from 0.35% to 2.44% of all CD45+ cells within 4 days after mRNA vaccination.

  • mRNA vaccination recruits and matures NK cells in the spleen.
  • Terminally mature NK cells increased from 0.85% to 3.91% of all CD45+ cells by 7 days post-vaccination.
  • Depleting NK cells before vaccination reduced serum levels of IL-1β and IL-6 shortly after vaccination.
  • NK cell depletion led to a greater than 5-fold reduction in Spike mRNA copies in draining lymph nodes.
  • Despite reductions in early inflammatory cytokines and Spike-specific T cells, NK cell depletion did not significantly affect Spike-specific antibody levels at 1 and 6 months post-vaccination.
