How a protein controlling iron storage affects silver nanoparticle-induced iron-dependent cell death in lung cells

Updated

Abstract

Silver nanoparticles (AgNPs) caused dose-dependent cytotoxicity and lung damage in both in vitro and in vivo models.

  • AgNPs exposure is associated with iron dysregulation through NCOA4-mediated ferritinophagy.
  • Excessive free iron from AgNPs leads to depletion of glutathione and increased levels of reactive oxygen species.
  • Alterations in ferroptosis-related proteins and lipid peroxidation were observed following AgNPs exposure.
  • Ferroptosis inhibitors significantly reversed the effects of AgNPs on lung toxicity.
  • Reduced expression of NCOA4 and autophagy inhibition decreased AgNPs-induced ferroptosis.
