High levels of MLKL cause liver cell death that leads to aging, ongoing inflammation, and scarring

Updated

Abstract

MLKL overexpression in hepatocytes led to a 4-fold increase in necroptosis and associated liver damage.

  • Necroptosis and cellular senescence are linked to increased inflammation with age.
  • Overexpression of MLKL resulted in elevated necroptosis and cell death in the liver.
  • Increased levels of p16, p21, and SASP-factors were observed alongside necroptosis.
  • Higher levels of proinflammatory factors and mononuclear cell clusters were detected in hMlkl-KI mice.
  • MLKL overexpression altered gene expression related to cellular senescence, inflammation, and drug metabolism.
  • The changes observed in 6-month-old hMlkl-KI mice were associated with increased liver fibrosis.
