Using network analysis and drug testing to find new insomnia treatment uses for Sishen Wan

Abstract

Sishen Wan (SSW) significantly reduced locomotor activity and prolonged sleep time in a dose-dependent manner.

  • SSW increased non-rapid eye movement (NREM) sleep in normal rats, indicating a solid sedative effect.
  • In various insomnia models, SSW improved sleep by increasing NREM sleep time and reducing sleep latency and fragmentation.
  • 5-HT was identified as a key target for the anti-insomnia effects of SSW.
  • Active compounds potentially responsible for SSW's effects include (S)-propranolol, nuciferine, zizyphusine, and N,N-dimethyl-5-methoxytryptamine.
  • The findings suggest that SSW may serve as a therapeutic option for treating insomnia.
