Changes in nerve cell receptors in the aging bone lining: From detailed cell maps to repair targets

Updated

Abstract

Seven major neuronal receptor families are identified in periosteal homeostasis, with 29 receptor genes actively transcribed in periosteal stem cells.

  • Neuropilin 2 (Nrp2) exhibits a progressive age-associated increase in expression from young to old mice.
  • The increase in Nrp2 is associated with a decline in specific progenitor cells and activation of stress-response genes.
  • Dysregulation of the axon guidance ligand Slit2 and downregulation of osteocalcin-associated genes are noted with aging.
  • Age-related periosteal dysfunction includes thinning of the cambium layer and impaired progenitor-immune interactions.
  • Neuronal receptor modulation strategies may provide a multimodal approach for skeletal regeneration and aging intervention.
